Keratinocytes are the predominant cell type of epidermis and form the outer protective barrier of the skin. Located primarily in the basal layer, these cells undergo controlled differentiation as they migrate through the stratum spinosum and stratum granulosum toward the stratum corneum.
Primary human keratinocytes are widely used in vitro to study epidermal biology, barrier formation, and cellular responses relevant to wound healing, inflammation, and UV radiation exposure. Because keratinocyte differentiation and keratin isoform expression are tightly regulated, reliable culture conditions are essential for physiologically relevant skin models.
